South Sydney have alerted the NRL Integrity Unit to a photograph of Latrell Mitchell which began circulating online on Monday.
In the image, Mitchell is seen leaning over a table on top of which appears to be white powder.
There is no suggestion what can be seen is an illicit substance.
On Monday afternoon, Souths confirmed the image was taken following a personal appearance by Mitchell in Dubbo at the weekend.
"The South Sydney Rabbitohs have been made aware of a photograph circulating through social media of Latrell Mitchell following a personal appearance in Dubbo on the weekend," the club said in a statement.
"The Club has informed the NRL Integrity Unit about the photograph.
"The Rabbitohs will be making no further comment at this stage."
According to Sydney Morning Herald, Souths are set to quiz the star fullback on the photo, and that the NRL has confirmed it has been made aware of the matter.
Mitchell is currently sidelined with a foot injury which ruled him out of NSW's successful Origin decider last month.
After a disastrous star to the year, the Rabbitohs made a mid-season surge towards an unlikely finals berth before a recent string of losses sat them back to 15th on the table.
