A 16-year-old boy has died in a quad bike crash in the remote Northern Territory community of Wadeye, while a 12-year-old boy and a 19-year-old man have been airlifted to Darwin.

The crash occurred about 11am on Wednesday, with an NT Police spokesperson saying the trio, along with another 19-year-old man, were all on the quad bike when it crashed.

Shortly after the accident, health clinic staff drove past the crash site near Manthathpe Bridge and took the injured to the local clinic, the spokesperson said, with initial reports suggesting "abdominal injuries and abrasions".

However, at 3:10pm, the clinic told police the 16-year-old's condition had "rapidly deteriorated", and he was declared dead a short time later.

"One of the 19-year-olds and the 12-year-old have now been conveyed to Darwin via Careflight for further assessment," the spokesperson said.

"The exact circumstances of the crash remain under investigation."

About 400 kilometres south-west of Darwin, Wadeye is one of the largest Indigenous communities in the NT. Detectives from the Major Crash unit are expected to arrive in the community on Thursday to continue investigations.
